The results of the research are intended to lead to societal impact and policy making to combat smoking related disease in the Netherlands.Īs a PhD Candidate at the Faculty of Health, Medicine and Life Sciences, you will be employed by the most international university of the Netherlands, located in the beautiful city of Maastricht. The project is financed by the Dutch Food and Consumer Product Safety Authority (NVWA) and is commissioned by the Ministry Of Agriculture, Nature and Food Quality (LNV). In this project, relevant read-out parameters will be investigated by using state-of-the-art molecular biology and imaging tools and in the end this should result in a PhD thesis. Subsequently, using in vitro work, cultures of neuronal cell populations will be used to investigate activation of relevant receptors and the effect of additives such as menthol. In these studies, we will also assess whether or not individual differences in nicotine metabolism in the mouth have an impact on these processes. First, we will determine the dose-range of nicotine in the mouth of users of cigarettes and other tobacco/nicotine products in human volunteer studies and will investigate the correlation with smoking behavior (such as puff frequency, puff volume etc.) and with activation of known reward centers in the brain (using fMRI). In the present project, we will investigate this hypothesis by a combination of human volunteer studies and in vitro work using cultured neuronal cells. Moreover, we postulate that levels of nicotine in the mouth influence smoking behavior and that the presence of additives (such as menthol) has a significant impact on this. ![]() We now hypothesize that, before inhalation, nicotine already binds to receptors present on free nerve endings in the mouth and nose cavity, which subsequently signal to reward centers in the brain. The current paradigm states that nicotine exerts its pharmacological effect by uptake in the blood via the lungs and subsequent transport to the brain. Nicotine is a highly addictive substance and triggers dopaminergic reward centers in the brain by binding to nicotine receptors in the peripheral and central nervous system. In addition to conventional cigarettes, alternative tobacco products or so-called nicotine delivery systems (vapes, e-cigarettes) are gaining popularity. Unfortunately, despite efforts to reduce the use of tobacco, currently 3 million people in the Netherlands still smoke.
